Determination of the Hepatitis C Virus Genotypes With‘Pyrosequencing’ Method

Abstract

Purpose: Determination of HCV genotypes in terms of management and treatment of chronic hepatitis C patients provides important information. In HCV genotype 1b, both lower rate of response to interferon treatment occurs and there is more risk for development of hepatocellular carcinoma. The purpose of this study was to investigate the distribution of HCV genotypes among patients from different clinics with the pre-diagnosis of chronic hepatitis C and positive for HCV RNA in blood samples.
Materials and Methods: Blood samples of 146 patients were included in this study. Ninety- eight patients were females (67.1%) and 48 males (32.9%). HCV genotype analyses of the samples were investigated by pyrosequencing method (Qiagen, Germany).
Results: Genotype 1, genotype 4 and genotype 2 were detected in 90 (61.7 %) of patients, 52 (35.6 %) of patients and 4 (2.7 %) of the patients respectively. In genotype 1 group, genotype 1b was detected in 77 of 90 samples and genotype 1a was detected in 5 of 90 samples 1, however subgenotyping failed in 8 patients.
Conclusion: As a result, HCV genotype 1b is the most common genotype in patients with chronic hepatitis C.


Hepatit C Virüs Genotiplerinin Pirosekanslama Yöntemi ile Belirlenmesi

Amaç: HCV genotiplerinin belirlenmesi kronik hepatit C.li hastaların takip ve tedavisi açısından önemli bilgi sağlamaktadır. HCV genotip 1b’de hem interferon tedavisine yanıt daha düşük oranda meydana gelmekte hem de hepatosellüler karsinoma gelişme riski daha fazla olarak bulunmaktadır. Bu çalışmada, kronik hepatit C ön tanısı ile farklı kliniklerden gelen hastaların HCV RNA pozitif kan örneklerinde HCV genotiplerinin belirlenmesi amaçlandı.
Gereç ve Yöntemler: çalışmaya 146 hastadan elde edilen kan örneği dahil edildi. Hastaların 98 (%67,1)’i kadın 48 (%32,9)’i erkek idi. Еrneklerde HCV genotip analizi Pirosekanslama yöntemi ile araştırıldı.
Bulgular: HCV genotiplerinin 90 (% 61,7)’ı genotip 1; 52 (%35,6)’si genotip 4; 4 (%2,7)’ü genotip 2 olarak bulundu. Genotip 1 olarak bulunan 90 örneğin 77’si genotip 1b; 5’inin genotip 1a olduğu belirlenirken, 8’inin alt tipi belirlenemedi.
Sonuç: Sonuç olarak, kronik hepatit C’si olan hastalarda en yaygın genotipinin 1b olduğu saptandı.
